What the medication is used for

Compounded semaglutide with B12 is a compounded medication. Compounded medications are not approved by the FDA, and the FDA does not review them for safety, effectiveness, or quality as compounded products.

A licensed provider may consider compounded semaglutide with B12 for weight management, based on your health history, current medications, allergies, location, and other clinical information. This page does not describe an FDA-approved product.

Who may not be able to use it

A provider may determine that compounded semaglutide with B12 is not appropriate if you or a family member has a personal or family history of medullary thyroid carcinoma, or if you have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). Animal studies of this class of medication have shown thyroid C-cell tumors, and this risk in humans has not been ruled out.

A provider may also determine it is not appropriate if you have a history of pancreatitis, severe gastrointestinal disease, a known serious allergy to any ingredient, or if you are pregnant, trying to conceive, or breastfeeding.

Important warnings and precautions

Compounded semaglutide with B12 carries warnings that a provider will review with you before prescribing, including a boxed warning about the risk of thyroid C-cell tumors seen in animal studies with this class of medication, and the related contraindication for people with a personal or family history of medullary thyroid carcinoma or MEN 2.

Other warnings include inflammation of the pancreas (pancreatitis), gallbladder problems, low blood sugar (which is more likely if you also take insulin or a sulfonylurea), kidney problems related to dehydration, vision changes, allergic reactions, gastroparesis and severe slowing of stomach emptying, and mood changes including depression or thoughts of self-harm.

Tell any surgeon, anesthetist, or endoscopist that you use this medication, because it slows gastric emptying and can affect procedures that require an empty stomach.

See Important Safety Information at /safety-information for the full safety summary that applies across Julora's compounded medications.

Common side effects

Common side effects reported with this class of medication include nausea, vomiting, diarrhea, constipation, stomach pain, indigestion, burping, tiredness, headache, dizziness, and injection-site reactions.

Important interactions

This medication can interact with insulin, sulfonylureas, and other medications that lower blood sugar, increasing the risk of low blood sugar. It can also affect how well other oral medications are absorbed because it slows stomach emptying.
